Investigation of interconnections between AgrA and σB regulons

OBJECTIVES:
The objective of this project under the joint supervision of UB and NUIG is to investigate the crosstalk between cell communication (mediated by AgrA) and stress response (mainly mediated by σB). The role of AgrA and have been studied for several years by UB and NUIG respectively. Recent evidence suggests that these regulators may interact in a way not previously expected. In this project we will investigate the crosstalk between these two regulators. Reporter strains will be constructed that allow the activities of the two regulators to be monitored by recording cellular fluorescence, either in the presence or absence of the other regulator. Using a combination of flow cytometry and fluorescence microscopy, the activities of these important regulators will be measured in both planktonic cells and in biofilm.

Catarina MOREIRA MARINHO
MY PREVIOUS TRAINING AND EXPERIENCE:
I’m from Portugal and I’m 25 years old. I hold a Bachelor degree in Genetics and Biotechnology and a Master degree in Molecular Genetics, both from University of Trás-os-Montes and Alto Douro, Portugal. Up to date, I’ve developed research on studying antimicrobial resistant bacteria and plant varieties using omic approaches, and published 6 papers, 2 book-chapters and several communications in scientific meetings.
WHY I WANTED TO JOIN THE PROJECT :
I applied to List_MAPS Programme because it is an ambitious project, aiming to obtain important results in my main research areas of interests. This project allows the integration of several vanguard techniques, headed by a team of renowned researchers studying Listeria. Also, the international background of a European project enables the contact with different research realities, making this project ideal for future follow-up of my academic and scientific career.
ABOUT MY RESEARCH PROJECT :
This project is under the joint supervision of UB and NUIG and aims to investigate the crosstalk between cell communication (mediated by AgrA) and stress response (mainly mediated by σB) on Listeria monocytogenes. Recent evidence suggests that these regulators may interact in a way not previously expected. Reporter strains will be constructed that allow the activities of the two regulators to be monitored. Using a combination of flow cytometry and fluorescence microscopy, the activities of these important regulators will be measured in both planktonic cells and in biofilm. The role of some ncRNA on the crosstalk between the two regulons will be ascertained. Mutants’ phenotypes will be assessed under specific environmental conditions.
